Because of the season-ending injury to Hagler, third-round draft pick in 2008 Philip Wheeler, is expected to see more time for the Colts.
Undrafted rookie Ramon Humber will also be expected to step up due to Hagler's absence.
Both Wheeler and Humber have yet to start a game for the unbeaten Colts (7-0), who will host the Houston Texans (5-3) Sunday.
Indianapolis will also promote rookie linebacker Cody Glenn from the practice squad to the active roster.
Hagler, in the midst of his fourth year with Indianapolis, has logged 26 tackles.
He has a total of 114 tackles and two forced fumbles in 37 career games.
Meanwhile, coach Jim Caldwell also announced Wednesday that no timetable was set for the return of cornerback Kelvin Hayden and WR Anthony Gonzalez, who are both dealing with knee problems.
